with what little free time I've had, I've managed to troubleshoot my way to what will likely be my primary setup... microflow LCMS. I've been flame-pulling fused quartz silica capillaries with inner diameters of 20-75 microns, down to 5-10 microns. this has improved the sensitivity of the signal by a factor of 1000. flowrates are anywhere from 10-50 microliters/min.

I've also been able to determine, because of my new-found stable spray configuration, that I
have polyethylene glycol contamination; I verified this using this, and the numbers match up within 1 mass unit.
This may be residual from the methanol I purchased, which is stored in a PE bottle, and DI water that was also stored in PE bottles.
So the moral of the story is, use glass whenever possible, for storing samples and solvents.

Once I get the contam situation under control, and a pre-column filter and column or two, I will set up a po box and begin accepting samples.

I bought an autosampler and another hplc stack, three pumps, which brings the total to four. I now have the option of running multidimensional liquid chromatography. I will now be able to run 48 samples at a time, including blanks and controls. will update soon after it arrives this weekend. I have the precolumn filter, and C18 columns, I need to get a phenyl-hexyl for proper 2D-LC.

I wish someone had FTIR, I would love to collaborate on analytical efforts.

Do you have reference standards or another way to be able to ID less known alkaloids in complex mixtures? I am thinking of Trichocereus TA's.
urtica is a fictional character. nothing written by this fictional character has anything to do with reality. if urtica was real, and performing any activities that are restricted by certain governmental forces, these activities would be performed in Heaven where nothing is true & everything is permitted.

before sending samples, make sure you note the following on a piece of printer paper:

nexus un
sample name (if plant material, species name)
if it is plant material, any tips on extraction methods would help, but isn’t mandatory

I was doing annual maintenance yesterday (cleaning ion optics and the trap), when the trap rolled off the bench, and crashed on the floor, shattering the quartz spacer rings Shocked

mass spec is down (for now).


I ordered a complete surplus analyzer to replace the entire assembly.. ion optics, ion trap, and electron multiplier, for $450. Hopefully, it will also address the mass accuracy issues I was having.
My coworkers were more bummed out than I was, because I knew I could just replace the analyzer. Analogously, it's like I busted a couple rods in a classic car engine. Rather than rebuilding, I just bought a new engine.

this entire assembly is the mass analyzer module. The quartz spacer rings are the two white objects separating the trap endcaps and the ring electrode, in the middle.
